How students should use this module

Read the molecular genetics background

Review the white-1 locus, Doc-retroposon insertion, sex-linked inheritance, PCR, and gel electrophoresis.

Perform DNA extraction

Work in groups, extract DNA from one fly type, measure sample concentration and purity, and record the values.

Set up the PCR reaction

Review the PCR cycler program, select the most suitable conditions, and prepare the reaction as instructed by the TAs.

Use the Virtual PCR Tool

Upload the five DNA sequence files, enter PF1, PF2, and PR3, run the required PCRs, and record fragment sizes.

Interpret PCR and gel results

Use 467 bp and 704 bp fragments to infer genotypes, phenotypes, heterozygotes, and expected gel bands.

Submit the assignment

Complete Assignment #8 and submit a physical copy or scanned PDF through Canvas.
